The globus pallidus (GP) is one of the main relay centers of the basal ganglia. It has a much higher dopamine concentration than any other brain region. Changes in the dopamine concentration can alter both the chemical coding as well as the epigenetic status of the neurons in the GP. In this study, we show DNA methylation levels are similar in the rat GP nucleus and the striatum. However, the distribution of the 5-hydroxymethylcytosine, a marker for 5-methylcytosine that is not present in mature neurons, was found to be lower in the GP than in the striatum. Moreover, the CpG content is higher in the GP and this correlates with high gene expression levels of genes encoding the transition factor P300. We also measured modifications of histone H3 and H4 at the promoter CpG islands of the CACNA1S (P/Q-type calcium channel alpha1 subunit) and LAMP1 (lysosome-associated membrane protein 1) genes, and found no difference between the GP and the striatum. However, a higher level of the actively transcribed (H3K4me3) histone modification was found in the GP. The findings indicate that the regulatory elements of these genes are similar, but that the transcriptional activity of these genes is higher in the GP.Q:

The intracellular distribution of protein kinase C was examined in situ by immunocytochemistry and immunoelectron microscopy in fixed cells. In human umbilical vein endothelial cells, immunostaining with two specific antibodies against non-phosphorylated and phosphorylated protein kinase C, respectively, disclosed considerable differences in the distribution of both types of protein kinase C in all the studied cell types. In aortic smooth muscle cells and in macrophages, no immunostaining for the non-phosphorylated protein kinase C was observed. The two protein kinase C-specific antibodies stained endothelial cells on the abluminal (outside) cell surface in addition to endothelial cells on the luminal (inside) cell surface. In the bovine retinal pigment epithelium, the two protein kinase C-specific antibodies stained the inner segment and the outer segment of the photoreceptor cells; immunostaining was also seen in the cells of the outer nuclear layer. A diffuse immunoreaction in neuronal processes was observed.
